Properties: This host strain is a derivative of the Hobby strain and can be used for bioengineering where purified products with reduced endotoxicity are desirable. Enterobacterial common antigen is no longer produced and a UDP N-acetylglucose epimerase has been chromosomally integrated increasing the availability of UDP-GalNAc. The O-antigen ligase has also been removed so that recombinant glycans are not removed from Und-P and attached to surface lipid. wzzB, the O-antigen chain length regulator of Escherichia coli, was removed and replaced by wzD-wzE, the chain length regulators from Streptococcus pneumoniae TIGR4, in the Hobby strain using CRISPR technology, in order to increase polymerization of plasmid expressed S. pneumoniae capsule glycans.
